Maleimide functionalized cyclooctyne derivatives for incorporation of cyclooctyne groups into sulfhydryl compounds or biomolecules. Cycloctene is useful in promoting Cu-free cycloaddition reactions to acetylene. This dibenzocycloctene will react with the azide-functionalized compound or biomolecule without the need for a Cu (I) catalyst to produce a stable triazole bond. PEG lipophilic spacers help reduce aggregation and precipitation problems when labeling biomolecules.
